A Simple Procedure for Quantitatively Measuring Pseudomonas aeruginosa Slime Production

چکیده

Pseudomonas aeruginosa is an opportunistic bacterial pathogen most associated with infection of the lungs of cystic fibrosis (CF) patients. It produces extracellular polysaccharides collectively referred to as slime that includes one specific virulence factor known as alginate. Current methods to measure the actual production of this slime require either expensive equipment, or they are not quantitatively accurate. This study reports a method that is simple and yields statistically reproducible results thereby enabling researchers to easily follow the rate of slime production in the bacteria. Ongoing studies will use this method to study the effect of growth media containing different components upon the production of slime. These studies may have in vivo relevance to the production of this virulence factor in the lungs of CF patients and in other infectious settings.

Alginate production by clinical nonmucoid Pseudomonas aeruginosa strains.

The slime material from a revertant nonmucoid variant, derived by serial passage of a heavily mucoid Pseudomonas aeruginosa strain isolated from a patient with bacteremia, was found to contain 16% uronic acids, 48.5% carbohydrates, 11% protein, and 2% lipids. Chromatographic analysis by ion exchange chromatography revealed that this extracellular material consisted of three fractions, one uroni...

Slime of Pseudomonas aeruginosa: in vivo production.

Indirect hemagglutination inhibition tests were employed to detect slime in concentrations as low as 1 mug/ml. Increasing concentrations of slime resulted in proportionately greater inhibition of hemagglutination. Peritoneal aspirates and plasma of mice injected with slime were shown to exhibit the inhibitory activity of slime. The rapid dissemination of slime into the peripheral circulation wa...

Antibiotic resistance in Pseudomonas aeruginosa is associated with decreased fitness.

The number of clinical specimens containing β-lactam-resistant Pseudomonas aeruginosa isolates is increasing. However, whether resistance is associated with reduced fitness is still uncertain in clinical Pseudomonas aeruginosa isolates. In this study, we aimed to determine whether β-lactam resistance conferred a fitness cost in Pseudomonas aeruginosa. Growth rate, extracellular slime production...

Post Irradiation Evaluation of Biofim Formation, Hydrophobicity, Antimicrobial Resistance, Alginate Alg D Gene of Pseudomonas Aeruginosa Recovered From Urinary Catheter of Cancer Bladder Patients

Colonization of biomaterials surface by Pseudomonas. Aeruginosa allows them form biofilms and be protected from host defense mechanisms and antimicrobial chemotherapy. Alginate production plays a central role in their pathogenesis. The research was to evaluate post irradiation effect on biofilm, hydrophobicity, resistance, alginate gene of Pseudomonas aeruginosa infection associated with urinar...

Comparison between batch and fed-batch production of rhamnolipid by Pseudomonas aeruginosa

This paper presents a comparison between batch and three different sets of fed batch fermentations forrhamnolipid production by Pseudomonas aeruginosa. The batch run was performed with 500 ml of culturemedium having the initial glycerol and sodium nitrate concentrations of 30 and 8.3 g/l, respectively. For a fedbatch run with nitrogen source in feed, 250 ml of the nitrogen exc...
